Tirzepatide is a 39-amino-acid synthetic peptide with a fatty-acid attachment. The fatty acid enables binding to blood proteins, extending circulation time and supporting a half-life of approximately 5 days — sufficient for once-weekly dosing protocols in research models. Once in circulation, it activates two receptors that the gut normally engages postprandially: GIP and GLP-1.
